چکیده

Transport of secretory proteins into the mammalian endoplasmic reticulum can bevisualized as a sequence of various steps which include membrane association, mem-brane insertion and completion of translocation. I t turns out that this transportdepends on the hydrolysis of nucleoside triphosphates at various stages: (i) There isa GTP requirement in ribonucleoparticle-dependent transport. This GTP effect isrelated to the GTP binding proteins signal recognition particle (SRP) and dockingprotein, (ii) There is an ATP requirement in ribonucleoparticle-independent trans-port. This ATP effect is related to the cytosolic (termed cis-acting) molecular cha-perone hsp70. (iii) Recently we addressed the question of whether there areadditional nucleoside triphosphate requirements in protein transport into mamma-lian microsomes. We observed that a microsomal protein which depends on ATPhydrolysis is involved in membrane insertion of both, ribonucleoparticle-dependentand -independent precursor proteins. The azido-ATP sensitive protein was shown tobe distinct from the lumenal (termed trans-acting) molecular chaperone BiP.

Molecular mechanism-based model to enhance outcomes of dietary intervention studies for disease prevention.

Advances in "omics"-based fields have produced an explosion of new information, fueling high expectations for improved public and individualized health. Unfortunately, there exists a widening gap between basic biochemistry and "omics"-based population research, with both disciplines failing to translate their full potential impact to human health applications. A paucity of comprehensive study s...

Yeast as a touchstone in post-genomic research: strategies for integrative analysis in functional genomics.

The new complexity arising from the genome sequencing projects requires new comprehensive post-genomic strategies: advanced studies in regulatory mechanisms, application of new high-throughput technologies at a genome-wide scale, at the different levels of cellular complexity (genome, transcriptome, proteome and metabolome), efficient analysis of the results, and application of new bioinformati...

Structural insights into the signal recognition particle.

The signal recognition particle (SRP) directs integral membrane and secretory proteins to the cellular protein translocation machinery during translation. The SRP is an evolutionarily conserved RNA-protein complex whose activities are regulated by GTP hydrolysis. Recent structural investigations of SRP functional domains and interactions provide new insights into the mechanisms of SRP activity ...

Challenges in enzyme mechanism and energetics.

Since the discovery of enzymes as biological catalysts, study of their enormous catalytic power and exquisite specificity has been central to biochemistry. Nevertheless, there is no universally accepted comprehensive description. Rather, numerous proposals have been presented over the past half century. The difficulty in developing a comprehensive description for the catalytic power of enzymes ...

COMPREHENSIVE STUDY OF SOME PLASMA BIOCHEMICAL PARAMETERS IN RELATIONTO BONE DISEASE IN HEMODIALYSIS PATIENTS IN ISFAHAN

The concentrations of serum calcitonin', parathormone (PTH), amylase, alkaline phosphatase (ALKP), calcium and phosphorus were studied in pre-and post-dialysis patients with chronic renal failure. All patients had extremely elevated serum urea and creatinine concentrations with the mean values of143.8 and 10.5 mg/dL respectively. The plasma amylase activity was higher than normal with a me...
